If it's in the form of a sign, I would use no punctuation. Capital letters might be another question. What I envision: Staff and Inmate Restrooms.

Hi,

The word 'inmate' suggest someone who is confined, eg in a prison or in a mental hospital.

In such a place, it seems unlikely that staff and inmates would share the same restrooms.
